Treatment with conditioned culture media with human mesenchymal stem cells on reserpine-induced thermal fibromyalgia model in rats

چکیده مقاله:
Background and Aims: Fibromyalgia (FM) is a chronic pain syndrome affected by the hippocampal function. Mesenchymal stem cells-conditioned medium (MSC-CMs) has shown antioxidant and neuroprotective potentials in treating various diseases. The present study aimed to investigate the efficacy of MSC-CMs on the thermal treatment of reserpine-induced FM and evaluate the concentration changes of tumor necrosis factor-alpha (TNF-α) in the hippocampus of rats under these conditions. Materials and Methods: 220 g. Animals received reserpine (1 mg/kg) in the FM groups, and subcutaneous injection of normal saline in other groups, once a day for 3 consecutive days. Then, the rats received MSC-CM (0.3 and 0.5 mg/ml protein content, intraperitoneally) once a week for three weeks. The thermal pain threshold of mice was evaluated using tail flick and hot plate tests at the beginning and end of the experiments. TNF-α concentration in the hippocampus was determined by an ELISA kit at the end of the experimental period. Results: The thermal pain threshold of rats in the FM group decreased significantly (P<0.01) in both pain tests compared to that of the control group. A significant increase (P<0.01) was observed in the level of TNF-α in the hippocampus of rats in the FM group compared to that of the control group. Treatment with MSC-CMs significantly reduced reserpine-induced pain and hippocampal TNF-a concentration despite the effect of fresh and conditioned medium on control rats. Conclusion: Treatment with MSC-CMs could control thermal pain in the rat model of reserpine-induced fibromyalgia by controlling the TNF-a level in the hippocampus.

Evaluating the Viability of Adipose Tissue-derived Mesenchymal Stem Cells in Diabetic Culture Media Model
Introduction: In diabetes mellitus, stem cells are exposed to inappropriate conditions such as an increase of glucose in extracellular space, oxidative stress, and deprivation from growth factors (following ischemia). This study was aimed to evaluate the effect of these conditions on stem cells. Methods: Stem cells were isolated from subcutaneous adipose tissue of rats. The cells were maintaine...
متن کاملStem Cell Conditioned Culture Media Attenuated Albumin-Induced Epithelial– Mesenchymal Transition in Renal Tubular Cells
BACKGROUND Proteinuria-induced epithelial-mesenchymal transition (EMT) plays an important role in progressive renal tubulointerstitial fibrosis in chronic renal disease. Stem cell therapy has been used for different diseases. Stem cell conditioned culture media (SCM) exhibits similar beneficial effects as stem cell therapy. The present study tested the hypothesis that SCM inhibits albumin-induc...
متن کاملBiochemical and morphological changes in bone marrow mesenchymal stem cells induced by treatment of rats with p-Nonylphenol
Objective(s):In previous investigations, we have shown para-nonylphenol (p-NP) caused significant reduction of proliferation and differentiation of rat bone marrow mesenchymal stem cells (MSCs) in vitro. In this study, we first treat the rats with p-NP, then carried out the biochemical and morphological studies on MSCs. Materials and Methods: Proliferation property of cells was evaluated with t...
متن کاملEffects of Bone Marrow Mesenchymal Stem Cells-Conditioned Medium on Tibial Partial Osteotomy Model of Fracture Healing in Hypothyroidism Rats
Background: Hypothyroidism is associated with dysfunction of the bone turnover with reduced osteoblastic bone formation and osteoclastic bone resorption. Mesenchymal stem cells (MSCs) secrete various factors and cytokines that may stimulate bone regeneration. The aim of this study was to determine the effects of MSCs-conditioned medium (CM) in hypothyroidism male rats after inducing bone ...
متن کاملThe Effect of Human Mesenchymal Stem Cells- Conditioned Media on Glioblastoma Cells Viability In Vitro
A novel target for cancer treatment is based on the effects of non-tumor cells, including hMSCs on tumor growth. However, the results are controversial: some studies showed that hMSCs inhibit tumor progression, while others found they promote tumor cell proliferation. In this study, we analyse the effect of human mesenchymal cells derived from umbilical cord tissue (hUC-MSCs) and bone-marrowmes...

Objective(s): The present day challenge is how to obtain germ cells from stem cells to treat patients with cancer and infertility. Much more efforts have been made to develop a procedure for attaining germ cells in vitro. Recently, human umbilical cord-derived mesenchymal stem cells (HUMSCs) have been introduced with higher efficacy for differentiation. In this work, we tried to explore the eff...
